Effects of starvation on the carnitine palmitoyltransferase of hepatic peroxisomes.

چکیده
I t is generally accepted that the mitochondria1 carnitine palmitoyltransferases play an important role in the regulation of hepatic fatty acid oxidation [ I ] . These two enzymes express their activities on either side of the mitochondrial inner membrane that acts as a barrier to the transfer of fatty acids into the mitochondrial matrix. The enzyme that lies outsidc this barrier is located on the mitochondrial outer membrane 121 and can be inhibited by physiological concentrations o f malonyl-CoA 131. An important aspect of the regulation o f the outer carnitine palmitoyltransferase is the change that occurs in its sensitivity t o inhibition by malonylCoA in different physiological and pathophysiological states. Thus, starvation 14-51 and diabetes [ 6 ] both cause a decrease in sensitivity t o inhibition by malonyl-CoA. It has been reported that a carnitine palmitoyltransferase activity is associated with isolated hepatic peroxisomes [ 71 and more recently it has been shown that this carnitine palmitoyltransferase activity can be inhibited by malonyl-CoA [8]. The purpose of this study was t o see how the activity and sensitivity to inhibition by malonyl-CoA of the carnitine palmitoyltransferase activity associated with isolated peroxisomes would be affected by starvation.

To elucidate the effect of a large dose of di (2-ethylhexyl) phthalate (DEHP), a plasticizer and peroxisome proliferator-activated receptor-α (PPARα) agonist, on hepatic peroxisomes, we orally administered 1,000 mg/kg/day, once daily, to 3 male and 4 female cynomolgus monkeys for 28 days consecutively. Light-microscopic and electron microscopic examinations of the liver were carried out in conj...

متن کاملThe role of changes in the sensitivity of hepatic mitochondrial overt carnitine palmitoyltransferase in determining the onset of the ketosis of starvation in the rat.
The relationships between the increase in blood ketone-body concentrations and several parameters that can potentially influence the rate of hepatic fatty acid oxidation were studied during progressive starvation (up to 24 h) in the rat in order to discover whether the sensitivity of mitochondrial overt carnitine palmitoyltransferase (CPT I) to malonyl-CoA plays an important part in determining...

The regulation of hepatic mitochondrial carnitine palmitoyltransferase-I (CPT-I) was studied in rats during starvation and insulin-dependent diabetes and in rat H4IIE cells. The Vmax. for CPT-I in hepatic mitochondrial outer membranes isolated from starved and diabetic rats increased 2- and 3-fold respectively over fed control values with no change in Km values for substrates. Regulation of mal...
